The present invention relates to compositions and methods for the effective lyophilization and reconstitution of virosomes.
Summary of the invention
[0016] The present invention provides biologically active compositions and methods for the preparation of highly lyophilization-stress resistant, hydratable virosomal lyophilisates and methods for their reconstitution. According to the invention, biologically active compositions refer to immunogenic compositions or pharmaceutical compositions comprising virosomes and a cationic lipid for effective lyophilisation and reconstitution of the virosome, and, in particular, to an immunogenic composition or a pharmaceutical composition, wherein a cationic lipid for effective lyophilisation and reconstitution of the virosome is present in the membrane of the virosome. Using the teaching of the invention, virosomes are obtainable which have superior lyophilization and reconstitution properties and which are particularly useful to deliver antigens, drugs and other pharmaceutical active substances including DNA, RNA or siRNA into cells. After lyophilization, they are still capable to deliver said substances to distinct cells through a targeting system, which recognizes surface markers of specific cell types, and, thus are specifically superior to other known delivery vehicles.
